    Acrylamide copolymer-based gel substrates with different viscoelasticity were employed to evaluate the viscoelasticity effect on the direct relation among cancer stemness and mesenchymal properties with induction of epithelial–mesenchymal transition (EMT) of human breast adenocarcinoma (MCF-7) cells in both normoxia and hypoxia. The softer gel substrate produced a large amount of surface molecule of cancer stem cells (CSC) marker CD44. In contrast, for the stem cell biomarker CD133 expression, their coefficient of damping (tan)-dependent manner was not contributed by EMT phenomenon and was an independent from acquisition of the EMT. The substrate damping as potential physical parameter emerged the important linkage to cancer stemness and EMT induction.

  • Cytotoxicity and potential application for regenerative medicineof of natural rubber latex particles

    We have successfully fabricated the cartilage/natural rubber latex (NRL) biocomposites via human mesenchymal stem cells (hMSCs) spheroid under hypoxic condition. It was revealed that NRL nanoparticles acted as the main component, providing surface heterogeneity for the spheroids and resulting in a mechanically stable structure with a modulus two times higher compared to controls. The formation of the cartilage/NRL nanoparticles biocomposite results in having a stiff property was demonstrated. The gene expression levels of SRY-box 9 (SOX9), aggrecan and type II collagen (Col-II), and formation of glycosaminoglycans were higher in spheroids under hypoxic conditions, and administration of NRL nanoparticles enhanced chondrogenic differentiation is successfully induced. The promise of NRL nanoparticles for well-controlled chondrogenic differentiation and mechanically stable cartilage tissue was demonstrated.

    We investigated poly (L-lactic acid) ( PLLA) substrate surface topography (fiber alignment) and the effects of different stiffness combinations were investigated. Mesenchymal characteristics in both normal and hypoxic conditions. In addition, we also studied the induction of epithelial-mesenchymal transition (EMT). CDDP treatment under hypoxia showed poor adhesion of MDA-MB-231 cells and marked suppression of E-cadherin (CDH1). A strong association between drug sensitivity and suppression of the epithelial marker for E-cadherin (CDH1) through substrate surface topography contributed to the anticancer drug resistance of MDA-MB-231 cells. PLLA substrates did not cause significant changes in EMT induction and acquisition, indicating that EMT does not affect drug sensitivity.

    Metastasis is one of the biggest issues in cancer treatment today. Mammary epithelial cells are supported in optimal by interaction with a soft matrix (microenvironment) with an elastic modulus of approximately 800 Pa1. On the other hand, after transformation, the breast tissue becomes progressively stiffer, and the tumor cells shrink significantly and become hypersensitive to the elasticity of the matrix. Additionally, and importantly, cancer cells invade blood vessels and enter the circulation during metastasis. The elastic modulus of fluids such as blood and mucus is approximately 50Pa, which is a very low stiffness. Therefore, it should be emphasized that the important association between cancer cell phenotype and changes in matrix stiffness is orders of magnitude smaller. This opinion focuses on the current understanding of cancer stem cells (CSCs) and epithelial-mesenchymal transition (EMT) in metastasis and identified the importance of research for engineered extracellular matrices with different viscoelastic properties to mimic the in vivo microenvironment.

    Metastasis is one of the greatest challenges in cancer treatment today. Normal mammary epithelial cells are optimally supported by interaction with a soft matrix (microenvironment) with elastic modulus of about 800 Pa. However, after transformation, breast tissue becomes progressively stiffer and tumour cells become significantly more contractile and hyper-responsive to matrix elasticity. In addition, importantly, the cancer cells penetrate into blood vessel and enter the circulation during metastasis. The modulus of fluid such as blood or mucus has very low stiffness of around 50 Pa. For this reason, the critical association between cancer cell phenotype and the change of matrix rigidity with an order of magnitude smaller should be emphasizing. This review highlights the current understanding of epithelial-mesenchymal transition and cancer stem cells in metastasis, and identified importance for investigation on artificial extracellular matrix with different viscoelastic properties, which is required to mimics in vivo microenvironment. The substrate damping coefficient (tan) as potential physical parameter emerged the important linkage to cellular motility, cancer stemness, and epithelial-mesenchymal transition induction. Although further investigation is required to clarify the efficacy of environmental stimuli (tan d) for tumors exhibiting stem cell-like properties, this review indicates that the cancer cells incubated on softer substrate might lead to express cancer stem cell biomarkers exhibiting high expression.

    When traction forces are generated in the cells underlying extracellular matrix (ECM) including artificial scaffolds, the cells feel essentially stiffness of the surrounding microenvironment and respond to applied forces and exert forces in the matrix, in which the traction forces can change cellular morphology and cytoskeletal structure. To date, analysis of cell morphology, including quantitative measurements such as cytoplasm roundness, cytoplasm elongation factor, nuclear elongation factor, ratio nuclear area to cytoplasm area ratio (AN/AC), nuclear dimension, and nuclear height, has been widely used in cancer diagnostics and hematology. Increasing evidence suggests that the extracted morphological features such as cell area and the length of major and minor axes, could also be used to analyze the dynamic changes of cells in diseases of the nervous system and cellular stress related phenomena. Furthermore, multivariate analyses of morphological data suggest that quantitative cytology may be a useful adjunct to conventional tests for the selection of new substances.
    Thus, understanding the interaction between microenvironment and cancer cells via cellular morphology is critical subject to tackle metastatic spread of cancer cells and its many associated issues. In this topic, the cellular morphological parameters and functional characteristics of cancer cells are summarized.

    白金を担持したアロフェンナノ粒子を水熱プロセスにより合成し,構造をXRD,FT-IR,TEM,化学状態をXPSにより調べた.水熱処理後に乾燥して得られた粉末のX線回折パターンはアロフェンの生成を示し,Pt成分の添加で変化することはなかった.一方でIRスペクトルには1300-1500 cm-1付近に新たなピークが現れ,アロフェンとPtとの間に形成された結合によるものと考えられる.XPSスペクトルの解析の結果,大気中,250℃で加熱後,Ptは一部PtOxの状態で存在するものの,主として金属状態となっていることがわかった.TEM観察により,数nmサイズのPt粒子が分散して存在していることがわかった.

    This Prize is awarded to individuals who have made valuable years of research or outstanding cumulative achievements that have contributed to the science or industry related to rubber. The prize was established in 1957 as a fund of $2,500 donated by his widow to The Chemical Society of Japan through The Society of Rubber Science and Technology Japan (SRSTJ) in 1957 to commemorate the late George Oenslager, an authority on the U.S. rubber industry who had taught rubber technology in Japan. Initially, it was awarded as a joint award by The Chemical Society of Japan and SRSTJ, from the first meeting in March 1958 to the 20th award in March 1996. After that, SRSTJ decided to select and present the award independently
